The technology, which shows strong potential for fighting cancer by enhancing one's immune system by increasing the number of cells that naturally destroy cancer, proved 100 percent effective in eradicating experimental human brain tumors implanted in mice.

OVERVIEW
A novel technology to treat cancer using combination gene therapy technology which both builds the body's immune system and destroys cancers simultaneously.

PRIMARY APPLICATION OF THE TECHNOLOGY
In Europe and the U.S., close to 20 million people live with cancer, and overall costs amount to $107 billion in the U.S. alone. Cancer gene therapy is expected to be worth $2 billion by 2005 and $3-5 billion by 2010.

HOW THE TECHNOLOGY SOLVES THE PROBLEM
The standard approach in utilizing gene therapy to combat cancer has been to attempt to replace defective genes in cancer cells, which has proven to be impractical because of the number of genes involved. The combination gene therapy uses GM-CSF and B7-2 to both build the body's immune system and destroy cancer cells. The treatment involves injecting the patient with two genes in one virus carrying a combination of B7-2 and GM-CSF.

The GM-CSF gene enters the patient's tumor and genetically alters the tumor so that it attracts the patient's antigen presenting cells (dendritic cells that capture antigens), which activate tumor-specific T-cells. The B7-2 gene also enters the patient's tumor and genetically alters it, so it can stimulate a larger number of, and stronger, T-cells to fight the cancer.

Combination Immunogene Therapy works by injecting two genes in combination into the patient's tumors directly to build the immune system and destroy cancer cells.

PATENT SUMMARY
The present invention provides two immunogene therapy protocols for the treatment of tumors. In particular, the present invention provides combinations of immune-modulating proteins that induce systemic immunity against tumors. In addition, the present invention provides humanized animal models suitable for the evaluation of anti-human tumor immunity and permit the identification of combinations of immune-modulating genes which when delivered to human tumor cells induce an effective anti-tumor response, including a systemic anti-tumor response.
